The BioCELL proFILE assay has been designed as an all-in-one real-time solution to probe mitochondrial and glycolytic energy metabolism at the same time. Using robust and accurate measurements of oxygen consumption rate (OCR) and proton efflux rate (PER), we are able to provide our clients with a complete picture of cellular energy metabolism for advanced cellular metabolic profiling. By using improved reagents that have minimal off-target effects, we profile glycolytic and mitochondrial energy metabolism of the same cells in the same wells, limiting the need for multiple assay tests providing a more cost effective solution for complete cellular metabolic profiling.

Using advanced data analysis tools we are able to provide deeper insights into the bioenergetic function of your cells including cellular ATP demands, capacities and flexibility. We use OCR and PER data to calculate rates of ATP supply from both mitochondrial and glycolytic pathways. Our data analysis provides an overview of the energetic phenotype of the cells and allows for a quantitative comparison of mitochondria and glycolysis in basal and max states.

Because ATP production rates are indicative of cellular activities (e.g., activation, proliferation, differentiation, etc.) our assay has broad applications. It can be used for assessing metabolic phenotypic changes and reveal pathway contributions during cell activation, proliferation, or differentiation. The assay can also be used to assess or screen for compound effect on cellular ATP production with a view of pathway impact.

  • This assay works for any cell line or primary cell type that uses both mitochondria and glycolysis to supply ATP. We can offer access to a range of different cell types including HEK293, HepG2, INS-1E, MCF-7, C2C12, PBMCs, T-lymphocytes, monocytes and macrophages.

  • This assay uses an improved uncoupler called BAM 15, which is mitochondrial specific and has a broader concentration range. This allows us to minimise optimisation but more importantly BAM 15 prevents interference with PER, so measurements of PER can be used accurately post BAM 15 addition to establish compensatory glycolysis with rotenone and antimycin a. We also include monensin, an ionophore that increases cellular ATP demand through increased activity of Na+/K+ pumps, which provides a more accurate measure of maximal glycolytic rate in certain cell types.

  • In some cases we would recommend optimising the assay, for example if a specific cell type or cell model is being used. We will also support any optimisation using our BioCELL optiMIZE assay service.

  • Yes. We run a post-hoc fluorescent based nuclear stain cell density test. This is key to normalise any cytotoxic effects of compounds that might skew the results. Whereby this test is not suitable for the cell model being used we can also offer total protein or dsDNA for normalisation.
