BioCELL mitoTEST
Our BioCELL mitoTEST is based on the widely recognised XF Cell Mito Stress Test. This well accepted assay is the gold standard for assessing mitochondrial function with intact cells in real-time. The assay provides key insights into mitochondrial function and is used frequently to investigate functional differences among cell types, drug candidates, and genetic or biochemical interventions.
How our test works.
Gain a comprehensive view of mitochondrial function.
The BioCell mitoTEST examines key mitochondrial respiratory parameters, including basal respiration, ADP-phosphorylation, maximal respiratory capacity and spare respiratory capacity. Spare respiratory capacity is used to evaluate the cells ability to meet an energetic challenge as cells go through activation, differentiation and/or proliferation, and has been implicated in cell fate determination.
Key parameters are examined using well established mitochondrial modulators.

Our protocol design involves 3 serial injections of oligomycin, uncoupler (e.g FCCP or BAM15), and rotenone/antimycin A. We can also assess acute effects of a test of compound on mitochondrial function by injecting this prior to injection of each mitochondrial modulator.

This assay works for any cell line or primary cell type that has functional mitochondria. We can offer access to a range of different cell types including HEK293, HepG2, INS-1E, MCF-7, C2C12, PBMCs, T-lymphocytes, monocytes and macrophages.

Although measurements of proton efflux rate will be assessed simultaneously, this test is designed to profile mitochondrial function. If interested in glycolytic function then see our BioCELL glycoTEST or our BioCELL profileTEST, which have specifically been designed to profile glycolytic function.

In some cases we would recommend optimising the assay, for example if a specific cell type or cell model is being used. We will also support any optimisation using our BioCELL optiMIZE assay service.

Yes. We run a post-hoc fluorescent based nuclear stain cell density test. This is key to normalise any pre-treatment that might skew the results. Whereby this test is not suitable for the cell model being used we can also offer total protein or dsDNA for normalisation.
